Writing methods section

In general, method description for imaging should include at a minimum:

  • the microscope manufacturer and model,
  • the objective specifications,
  • excitation and emission details (e.g. laser wavelength, filter set specs, confocal detector settings, etc),
  • capture settings (e.g. camera exposure or detector speed).

Most instrument specifications can be found in documentation on NUCore, or in the image metadata.  Including the absolute laser power at the objective is considered a gold standard for reporting on methods; the usage of a laser power meter can determine the power.
